Spot formation within ELISPOT assay wells

This image shows examples of spots formed within ELISPOT wells. Note the existence of both red and blue spots in this example. The spot colors are determined by the chromogenic substrates used. These are in turn linked to specific secretions using the appropriate detection antibodies.

The well on the right was produced by a double-color ELISPOT assay. As the name implies, this well illustrates the chromogenic footprints produced by two different types of cellular secretions (e.g. cytokines).
